Detecting The Expression Of The Autophagy-related Gene In The Nude Mouse Model Of Human Lung Cancer Cell Line A549

Objective The aim of the study is to establish the nude mouse lung cancer model by injection,and discuss the relationship between autophagy and tumor growth by detecting autophagy associated gene after the application of anticancer drugs.Methods Human lung adenocarcinoma cell line A549 was applied,took right anterior axillary injection in nude mice by different numbers of cells, according to tumor growth and survival of nude mice,chose the best number of cells to established tumor model in nude mice. Groups using different doses of anti-cancer drugs cisplatin and placebo,took observation on tumor growth, killed nude mice,recorded the weight of the tumor,extracted the tumor cells, applicated RT-PCR detection of beclin-1 gene expression.Results To establish the nude mouse lung cancer model by injection, the optimal concentration of A549 cells was 5×10~6个/0. 2 ml,and autophagy negatively correlated with tumor growth.
