| Fructus Gardeniae belongs to the Gardenia jasminoides Ellis. Its dry fruits were commonly used as Chinese herb drug. Previous investigations indicated the main chemical component was geniposide of iridoids. On the other hand, it exhibited several potent pharmacological activities, including antipyretic analgesic and liver-protecting activities and so on. In this study, the quality specification was established and the pharmacokinetics of geniposide in rats was systematically investigated, which should be helpful for acquiring a deep development and application as well as rationalizing the future drug design.1. The preparation of the reference substance and its determination Geniposide has been isolated from Gardeniae with gel column-chromato-graphy and purified by recrystallization. Its structure was identified with UV, IR and MS. A reversed-phase HPLC method has been established to determine geniposide in Gardeniae. The mobile phase was methanol-1 % acetic acid (30:70, v/v), and the UV detection wavelength was set at 238 nm. The results showed the contents of geniposide of 10 sources of Gardeniae were different with various producing areas. And the mean content was more than 4.0 %(g/g).2. The pharmacokinetics of geniposide in ratsA rapid and sensitive liquid chromatography method has been developed to determine geniposide in rat plasma. Geniposide and paeoniflo -rin (an internal standard) were extracted from plasma using protein precipitation with acetonitrile, then separated on a Diamonsil C18 column. |
