Primary Studies On The Strains And Their Metablic Products Of Endophytic Fungi From Camptotheca Acuminata Decne.

Camptotheca acuminata Decne.(CA), an important medical wood, contains many antitumor alkaloids. Some researches show that endophytic fungi in medical plants can produce similar physiological active products to the plants. So researches on the endophytic fungi and their products have been a new way to find medical resources.Fourteen endophytic fungi are isolated from CA. The basic physiological experiments of three strains: a8, j1 and g13 have also been done, which include studies on the culture temperature, initial pH value of the medium, carbohydrate and nitrogenous nutrition, the concentration of carbohydrate, nitrogenous nutrition and the whole components and some vitamins.The way to detect alkaloids from the culture of microorganism has been found. We utilize five alkaliodal precipitants in the screening of the ferment extracts: Drangendoff s reagent, Mayer's reagent, Wagner's reagent, Sonnenschein's reagent and Bertrand's reagent, together with one spray reagent: Modified Dragendoff s reagent. Alkaliods in the ferment extracts can be detected through precipitate reaction in test tube and color reaction on thin layer chromatography(TLC).The method to extract and isolate water-soluble alkaloids from the culture of Strain a8 has also been found. Extraction with diluted acid followed by adding two volumes of ethanol can result in a crude extract. Then two isolate methods have been tried: Amonium Reineckate Precipitation Method and Ion Exchange Resin Method together with silica gel absorption chromatography. We get two crystals with the...
