Objective:To investigate molecular genic feature of rotavirus in 210 children with enteritis in Kunming, China from 2002 to 2004.Methods:Stool samples were collected from 210 children who were admitted to the department of pediatrics, the first Affiliated Hospital of Kunming Medical College from September to December from 2002 to 2004. Polyacrylamide gel electrophoresis(PAGE) was used to detect rotavirus genic RNA and identify electropherotypes of group A rotavirus. Reverse transcription polymerase chain reaction(RT-PCR) and nest-PCR was performed to identify rotavirus G serotypes. RT-PCR was used to amplified NSP4 , the resulted cDNAs of 30 specimens were sequenced.Result: 143(68%) of 210 samples investigated were group A rotavirus gene positive by PAGE, and their electropherotypes were long types, none was short electropherotypes or mixed electropherotypes. RT-PCR were performed for 143 fecal specimens and VP7 gene products were obtained from 134(93.7%) samples. Net-PCR showed serotype G3 accounted for 54.5%(73) of these samples, serotype G1 accounted for 28.4%(38) ,co-infected by 2 or 3 rotavirus with different G type accounted for 6%(8),only four G4 was detected and 11 were nono-typable,none was G2 or G9.
