Objective: To produce a hierarchical model of rats' contusion brain injury and investigate the effects on the recovery of neurological functions and the expression of NGF after NSCs transplantation through intravenous injection.Methods: The rats' brain were injured by Feeney percussion device made by ourselves through the weigh drop(50g) from different height(10cm,20cm,30cm) . We observed the changes of neurological functions and histopathology made by different height 24hs post-injured and make sure the parameters used in the device. Make a middle degree model of contusion brain injury and GFP transgenic mouse derived NSCs were vein transplanted into the injured rats 24hs post-injured. One week later, we make an evaluation on the neurological functions and the expression of NGF by immunohistochemical stain to study the effects on the recovery of neurological functions and the expression of NGF after NSCs transplantation.Results: There was an obviously disturbance of neurological functions and changes of histopathology at the level of injury (50g×20cm ,50g×30cm). A significant difference in the evaluation of neurological motor function and cognition function between pre-injury and 24hs post-injury could be seen, and there was a low death rate at the level of injury(50g×20cm). Following NSCs transplantation two weeks later, the NSCs could be seen in the cortex surrounding the wound cavity.
