Purpose: To elucidate the structural evolution in Actinomyces viscosus biofilm formation and the spatial distribution of exopolysaccharides(EPS) in it. To comparatively analyze the change curve of water-soluble exopolysaccharides and water-insoluble exopolysaccharides in the biofilm formation , then further investigates the functions of the two different exopolysaccharides on adhesion, development, and maturation of Actinomyces viscosus biofilm.Materials and methods: 1.The Actinomyces viscosus biofilm wasmade by allowing bacterium to attach to the clean glass surface. The biofilm structures at 9,12,15,18 ,24 hours and exopolysaccharides distribution were visualized with fluorescent staining, BoBoTM3 and CalcofluorTMrespectively by confocal laser scanning microscopy (CLSM). 2. The biochemical method was applied to measure the production of soluble and insoluble exopolysaccharides by adhesive Actinomyces viscosus and planktonic ones in different culture times. Results: 1. Actinomyces viscosus biofilm can be approximately divided into three stages —initial adhesion (about 12 hours ago) , development (about...
