Background: Adenoid cystic carcinoma is one of the most common malignant rumors in the maxillofacial region ,metastasis is the essential character causing mortality from such tumor and little is currently known about mechanisms underlying it. FAK in the signalling pathways have been shown to regulate a variety of cellular functions such as cell spreading , migration , cell proliferation , apoptosis and cell survival. Up to now ,however ,there isn't any report about the role of FAK in Acc-M cells, and whether it has the same or similar effect on this kind of cell lines as in other tumors is still unknown .Objectives: In order to investigate whether FAK takes part in the intracellular signalling pathway involved in the biological characteristics of in vitro cultured high metastatic cells of adenoid cystic carcinoma(Acc-M),we selectively suppress the expression of FAK by using antisense oligodeoxynucleotides (ODNs).Materials and Methods: FAK oligodeoxynucleotide (ODN) was transfected into Acc-M cells with lipofectamine 2000. The difference in expression of FAK between the transfected and non-transfected cell lines was detected by RT-PCR and was observed by the immunofluorescence.Then by use of transwell cell culture chamber and wash way method, the difference in invasion and adhesion ability between the transfected and non-transfected cell lines was tested. MTT method was adopted in finding the change of proliferation. The effects of inducing Acc-M cell apoptosis were examined by Tunel and detected by flow cytometry.
