| Objective To evaluate bone marrow stromal cells as seeding cells and bio-derived bone as scaffold for bone tissue engineering though studying growth features of rabbit BMSCs adhering to bio-derived bone in vitro.Methord The method of density centrifuge was employed to purify bone marrow stromal cells, which was got from rabbit. The purified cells were cultured in vitro. After 4 passages of subculture, BMSCs were induced into osteoblast by osteogenesis supplement and seeded onto the bio-derived bone scaffold, which was derived from the spongy bone of rabbit. The growth features were observed under reverse phase difference microscope and scanning electro microscope respectively.Results BMSCs could easily be purified by the method of density centrifuge and cultured in vitro. After induced by osteogenesis supplement, BMSCs exhibited osteogeneric capability. The induced BMSCs adherented onto bio-derived bone and kept growth activity.Conclusion BMSCs are ideal candidate seeding cells for bone tissue engineering, and the bio derived bone loaded with BMSCs may be used as a scalffold on bone tissue engineering . |
