This work mainly focused on extraction and characterizations of rice bran polysaccharide (RBS), the antitumor activity both in vitro and in vivo of crude RBS were studied. The main results were shown as follows:1. The optimal extraction processing parameters for microwave-assisted extraction were the ratio of water to sample 10:1(w/w), microwave power 600W, extraction time 10 mins, and the yield 1.1%, compared with that of the hot water extraction method 0.7%. Pre-cooking was good for higher yield when amylase and cellulase were used for extraction. Three enzymes were used in the extraction and the highest yield was 1.8%.2. RBS from six extractions were hygroscopic, containing a little protein, no starch. Preliminary conclusion was that they were kinds of α-glucan. Different extraction influenced the structure of BRS.3. The RBS extracted by flavorzyme and microwave showed potent antitumor activity on S180 in vitro and the inhibition rate of S180 were 50% and 60%,respectively. High dosage of RBS by flavorzyme showed potent antitumor activity on tumor-bearing mice, and the highest inhibition activity of S180 were 56.9% at the dosage of 150mg/kg·d. RBS could improve superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and decrease malondialdehyde (MDA) in blood serum and liver homogenate in mice.
