Plant polysaccharide is an active area as functional substances. The extraction technologies of polysaccharide of Basella rubra.L were developed by using hot water ,and the ultrasonic sonic method, which provide with a base for the research on the function of extrcted polysaccharide in anti-tumor,anti-oxidation and anti-weary.The main results obtained are bellowing:1.Extraction rate: Influence of the factors such as solid-liquid ratio,the extraction temperature and the extraction duration on the yield of the polysaccharides was investigated. Experimental results indicated that the extraction time and the extraction temperature would significantly affect the extraction rate. Unter the present conditions,in hot water extraction, the optimum solid-liquid ratio was 1:55, the extraction time 6h with temperature at 75℃,and the extraction rate of 4.01% was obtained. In ultrasonic sonic extraction, the extraction rate of 17.02% was reached by using the wave capacity of 160w, at 70℃for 120min, which is 4.25 times higher the that of the hot water method,indicating the ultrasonic sonic technique is more applicable.2.Islation and purification: The DEAE-Sepharose-FF ionic exchange chromatographic method was applied to purify the rude polysaccharide, and six components obtained. The optional isolation conditions were established as: the column loading 120mg/10ml, the elution speed 1.6ml/min. the NaCl concentration gradient (0~0.6 mol/L) and the Tris-HCl buffer solution(pH7.6) were used to solve. Each components of polysaccharide was determined by phenol-sulfuric acid colormetry, suggesting the active material may be glycoprotein or proteoglycan.3.Function in anti-oxidization: Based on the AP-TEMED system and (1,10-phenanthroline) -Fe2+ system, the effect of the rude basella rubra polysaccharide on clearing oxygen radical, including superoxide anion radical and hydroxyl radical were studied. And the on inhibiting the H2O2 induced red blood cell hematolysis action was investigated. In present research, the maximum clearing rate of O2- ? and·OH were 54.32% and 52.94% respectively, and inhibition rate of red blood cell hematolysis was 55.76%.4.Function in anti-immunity, anti-tumor and anti-weary: Research of the rude polysaccharide affecting the small mice's immunity ability with the carbon grain puin experiment and the immunity organ weight-influenced experiment, and anti-tumor with S180 animal test model were conducted. The influence of the polysaccharide on mice's anti-stress experiment was tested too. The results implied that the polysaccharide with proper concentration had functions in all the aspects tested, throwing a light for futuer research.
