Solvianolic Acids which is the water-soluble effective part extracted from the roots of Salvia Miltiorrhiza Bunge have prominent pharmacological effects.But impurity components such as amylase,protein and tannins reduce Solvianolic Acids content and pharmacological effects. Ultrafiltration method was used in this paper to increase Solvianolic Acids Content and impurity components were removed meantime.After that cross-linked dextran gel was synthesized to separate and prepare pure component of Solvianolic Acids though HPLC method and the results were as fellows:1. It is obtained after study on analysis of content tests of components that light prevent method should be used when sodium nitrite-aluminum nitrite colrimetry was used to test Solvianolic Acids content.Concentration 0.2g/mL is the best concentration sodium chloride and iron trichloride method was used as method for measurement of tannin-removed effect when potassium permanganate method was used to determinate tannin content.Anthrone- sulphuric acid colrimetry was selected to determinate total sugar content after two total sugar content test methods were compared.2. Separation through ultrafiltration method to increase Solvianolic Acids content and correlated elements were tested. The membranes with MWCO:200,1500,3000 were tried and membrane with MWCO 200 was selected. The influence of various elements effecting T and the rate of Solvianolic Acids filtration were investigated, and the optimum conditions were confirmed as fellows: Concentration of water soluble part of Danshen 2.2 mg/mL,operational temperature 40℃and operational pressure 1.0 MPa,T and the rate of Solvianolic Acids filtration were 8.80and 2.47%.T and the rate of Solvianolic Acids filtration increased individually to 14.76 and 12.65% by useing adding water method. Impurities can be removed efficiently and the Solvianolic Acids content was increased.3. Cross-linked dextran gel was synthesized though liquid-liquid dispersal and cross-linking method.Dichloroethane was selected as oil phase.L18(37) orthogonal experiments were used to optimize effective elements:reacting time, churning speed,content of sugar solution, NaOH dosage, ratio of oil phase to liquid phase,1-chloro-2,3-epoxypropane dosage and PVAc dosage which affect average particle size and average pore size.It is obtained that average particle size was affected most notablely by PVAc dosage while average pore size was affected most notablely by 1-chloro-2,3-epoxypropane dosage. Suitable packings with average particle size 7.3μm and average pore size 26nm were selected as packings for HPLC separation though altering PVAc dosage and 1-chloro-2,3-epoxypropane dosage.4. Two components were confirmed to obtain and suitable separation temperature 20℃, eluent pH2.44 were selected though analysis HPLC experiments.Suitable eluent linear flow rate 4.0mL/min and concentration over loading method 3mL 5% were selected though preparative HPLC experiments.Two components were obtained with contents 80% and 90% individually under the optimum liquid chromatography conditions described as before.
