Radix Salviae miltiorrhizae is the Chinese traditional medicine of the heart cerebralparoxysm. In recent years, a great deal of pharmacology researches discover that thewater-soluble composition (salvinolic acids) of Radix Salviae miltiorrhizae have a goodapplication foreground for heart and cerebral paroxysm treatment. In order to clarify thechemical foundation of the water-soluble composition, the paper describes the preparation,isolation and chemical characterization of the salvinolic acids from Radix Salviaemiltiorrhizae.The70%enthanol crude extracts of Radix Salviae miltiorrhizae were screened by meansof combined HPLC-PDA-UV technique. By means of extraction and industrial preparativechromatography technology,70%enthanol extracts led to isolation of nine compounds. Thestructures of them were characterizated by MS, IR and NMR. They are rosmarinic acid (W1),lithospermic acid(W2),7”-methyle lithospermic acid(W3), salvianolic A(W4), salvianolicB(W5),danshensu(W7) and protocatechualdehyde(W8). W6and W9are two new compoundsnamed lithospermic acid C and4-(3,4-dihydroxy phenyl)-2-butenoate, which are firstlyseparated from Radix Salviae miltiorrhizae. Their molecular weights are495and195respectively.On the base of the analysis date on C18chromatographic column,the paper firstlysummed up the retantion behaviour of five salvacnolic acids on silica gel,sephedax LH-20and CG161chromatography. The salvianolic acids were five compounds of W1~W5.Except the analysis process by using CH3CN-H2O, other isolation processes usedCH3OH-H2O as the mobile phase. The results suggested that the flow sequence wasW1>W2>W5>W4>W3on the C18column, W1>W2>W4> W3>W5on the silica column, andW1>W4>W3>W2on the sephedax LH-20column. On the basis of the theory of “similarityand intermiscibility”, we conclude that the flow sequence was W1>W4>W3>W2on theCG161column. In summary,the five salvianolic acids followed that the flow discipline wasre-reserve HPLC on silica gel, sephedax LH-20and CG161column. It is noteworthy that there-reserve HPLC dicipline on silica column is maybe relate to the intermolecular special interation. The paper concludes the retantion behavior of salvianolic acids in chromatographypreliminarily, And a further research is needed.
