It is well known that deoxyribonucleic acid (DNA) is the transference base of information in the system of life. The destruction of DNA may cause the handicap of life, even discontinuity. Much attention is paid to the research of the interaction of DNA with other molecules in recent years. The Interaction of DNA with small molecules, especially drug molecules, may affect physiological and physical chemical character of DNA and change the transfer and copy of DNA. Hence, it is significant to research the interaction of DNA and small molecules in the promotion for understanding the interaction modes of DNA and protein and anticancer mechanism for human being, as well as to design and synthesize new drugs. DNA modified electrode shows a comprehensive application prospect in the relative research fields basing on its some special advantages, e.g. less sample consumption and higher sensitivity etc.In this thesis, we provide a general introduction and a review of the research on the interaction of drug with DNA. The interaction mechanism of rutin metal complexes with DNA and the catalytic oxidation of rutin-SAM-modified gold electrode to DA are also investigated.This thesis is divided into five parts:Chapter 1 General introduction and reviewThe development in the research of the DNA and bioactive molecules, as well as the basic composition of DNA, the interaction of DNA with the other substances and the research methods are reviewed herein, and the important theoretical and practical significance of the research of the interaction of DNA and the bioactive molecules in the pharmic filtration and biochemistry are also discussed.Chapter 2 The research of the synthesis, characterization and electrochemical behavior of rutin metal complexesThe rutin complexes with Cd(II), Co(II), Ni(II), Cu(II) and Zn(II) are synthesized herein, and are characterized by UV-vis, FT-IR, 1H NMR, fluorescence spectra and electrochemical methods, and the structure of these complexes is also confirmed. Chapter 3 The interaction of rutin transition metal complexes with DNAThe interaction of rutin transition metal complexes with CT DNA are researched by UV-vis, fluorescence spectra, viscosimetry and electrochemical methods, and intrinsic binding constants are calculated. It is proved in this experiment that rutin metal complexes bind with DNA in intercalation.Chapter 4 The electrochemical characters of rutin Ce3+ complex and the interaction with DNAThe interaction of rutin and Ce3+ in solution, as well as that of rutin Ce3+ complex and DNA, is investigated by electrochemical methods and UV-vis. It is proved that the complex of Rutin-Ce interacts with DNA in intercalation.The number of binding site is 1 and the binding constant K is 8.58×105 M-1.Chapter 5 The analytical applications and electrochemical behaviors of DA on the self-assembly modified gold electrodeThe stable rutin modified gold electrode is obtained by the method of mercapto self-assembly, and its electrochemical behaviors are investigated. It is proved that the electron transfer coefficient is 0.4, and the standard rate constant is 55 S-1. It is also found that rutin on the modified gold electrode may have redox reaction. The electrocatalystic oxidation reaction of DA on the rutin self-assembly modified gold electrode is researched by cyclic voltammetry and differential pulse voltammetry. The above experiments prove that rutin of the modified gold electrode has the ability to effectively catalyze DA.
