In this paper, the extraction technology of rutin from sophora japonical is optimized using response surface analysis.And the synthetic technology of chromium and rutin is optimized by orthogonal experimental design method.The combination of rutin and chromium is verified by Infrared spectroscopy,X-ray diffraction spectroscopy and Inductively coupled plasma atomic emission spectrometer. It is found that a supplementary role in lowering blood sugar of the combination on streptozotocin (STZ)-induced Wistar rat model of high blood sugar,through animal experiments.The main conclusions are as follows:1. Based on the single-factor experiment, the extraction technology of rutin from sophora japonical is optimized using response surface analysis.The effects of volume, time, the ratio of fluid-solid (mL:g), on the yield of rutin were studied and the parameters for optimum yield were obtained by using RAS softwave.The results showed that the optimum parameters is:71.23 percent ethanol volume, 2.2min,32:1(mL:g). In this situation, the extraction yield of rutin is 23.25%.2. The synthetic technology of chromium and rutin is optimized by orthogonal experimental design method, the effects of reaction time, temperature and mixing speed. The optimum process parameters were added anhydrous sodium carbonate,after rutin dissolved in ethanol, after 80 min added chromium chloride, The reaction time is 240 min, temperature is 70℃, stirring speed is 2100r/min.3. The combination of rutin and chromium is verified by Infrared spectroscopy,X-ray diffraction spectroscopy and Inductively coupled plasma atomic emission spectrometer.It is found that the reaction has generated a combination of chromium and rutin. It is found that the rutin amorphous changed by contrasting X-diffraction spectrum diagram of rutin and rutin chromium, indicating that the formation of a stable compound. Compared infrared spectra of diagram of rutin and rutin chromium, it is found that each functional peak has been displaced, and appeared Cr-O telescopic vibration absorption peaks in the place of 473.39 cm-1, which indicated the compound has Cr elements. The content of chromium is 0.6%, the combination of rutin and chromium measured by atomic emission spectroscopy. But the content is 3.8% after purification.4. It is studied the auxiliary lowering blood sugar function of the combination of rutin and chromium, by establishing hyperglycemia experimental animal models, rats as experimental subjects, the dosage referenced to other relevant research, in accordance with the "health food testing, and evaluation of new norms -2003". The results are as follows, there is significant difference between high-dose group of rutin chromium and the control group, low-dose group and the control group, positive drug group and the control group; there is not significant difference between high-dose and low-dose group, positive drug group and high or low -dose group. It is proved that the combination of rutin and chromium a supplementary role in lowering blood sugar of the combination on streptozotocin (STZ)-induced Wistar rat model of high blood sugar.
