OBJECTIVE To explore the effects of N2-l-Alanyl-l-glutamine on experimental acute intestinal failure in SD rats and compared to antibiotics.METHODS fifty healthy female SD rats were randomly divided into five groups:control group,model group,N2-l-Alanyl-l-glutamine group,Cefamandole sodium group,combined treatment group by N2-l-Alanyl-l-glutamine and Cefamandole sodium.Each group include ten rats.Control group with a sham surgery,other groups with a CLP surgery.Thirty six hours later after the CLP surgery,we examined the infection of abdominal cavity,leucocyte,the ratio of neutrophilic granulocyte,the serum concentration of lipopolysaccharide and phospholipase A2,and intestinal pathological changes.RESULTS(1) Thirty six hours later after the CLP surgery,model group's SD rats showed significant infection of abdominal cavity,part of intestinal putrescence,intestine conglutination,intestinal ulcer,and significantly lower in the leucocyte,the ratio of neutrophilic granulocyte,the platlet and the serum concentration of lipopolysacchari de and phospholipase A2 compared with control group(p<0.01).(2) Compared with model group,N2-l-Alanyl-l-glutamine group showed significantly lower in the infection of abdominal cavity,in the leucocyte,the platelet,ET(p<0.05),the ratio of neutrophilic granulocyte and the serum concentration of phospholipase A2(p<0.01).(3) Compared with model group,the Cefamandole sodium group showed significantly lower in the infection of abdominal cavity,in the leucocyte, the platelet,(p<0.05),the ratio of neutrophilic granulocyte and the serum concentration of lipopolysaccharide and phospholipaseA2 (p<0.01).And compared with N2-l-Alanyl-l-glutamine group,the above indite have no difference.(4) As expected,compared with model group,Combined treatment group showed significantly lower in the above indice(p<0.05).And compared with N2-1-Alanyl-1- glutamine group and Cefamandole sodium group,Combined treatment group showed significantly lower in the leucocyte,the ratio of neutrophilic granulocyte(p<0.05),the serum concentration of lipopolysaccharide and phospholipase A2(p<0.01).CONCLUSIONS(1) The CLP surgery could induce abdominal severe infection and result in intestinal failure in SD rats.(2) N2-l-Alanyl-l-glutamine is effective regarding treating intestinal failure induced by abdominal severe infection.And the effects is equal to antibiotics.(3) The combined treatment shows more effective than N2-l-Alanyl-l-glutamine or antibiotic alone.(4) The protective effects of N2-L-Alanyl-L-glutamine on severe infection induced intestinal failure may attribute to inhibiting lipopolysaccharide and phospholipase A2,the key mediators of inflammation,neutrophile infiltrating and inflammatory factor.
