Objective: To establish an animal model in goat by one-side Distraction osteogenesis ,and get primary knowledge of the evolution process of bone of mandibular regenerated by distraction osteogenesis. In the study, the expression patterns of basic fibroblast growth factor were studied using immunohistochemistry during distraction osteogenesis in goats.Methods: Ten goats(maleness,20-25kg)from the Taihang mountain were used in the current study . The goats were divided into five groups , which group contains 2 goats. operation was done using aseptic technique and with the animals under general anesthesia. A device for distraction used for people was fixed between molar and mandibular branch. After 3 days , latency period , the dist ractor was initiated at the rate of 1 mm every day , totally 10 days. When distraction completed , and the goats sacrificed after 0, 2 ,4 ,6 ,and 8 weeks , CR-X ray films of one side were taken respectively ,and new bone tissues were obtained, and observed with scanning electron microscpy and paraffin slices were made ,HE and immunohistochemistry stains of bFGF were completed on the slices.Results :During the experiments , the all goats were died because of anesthesia and operation .mandible midline obviously deflected to the right side. From a outside observation , the osteotomy site was filled with fibrous tissues in the distraction period, and new bone tissues begun to form, which couldn't be distinguished by our eyes after 2 weeks. From the X-ray film, we can see the gray scale continue to increase with time, after 8 weeks, the gray scale of new bone was similar to common bone . HE shows Tthat after the distraction , the gap was filled with fibrous clot. The mesenchymal cells in the fibrous tissue were observed. the trabeculae were formed in microcolumns parallel to the distraction force at 2 weeks .after 4 weeks in the consolidation period, the trabecular bone became mature comparally, and cartilaginous tissue was formed in the middle of the distraction callus. At the sixth week, resorption cavities were observed ,and After 8 weeks in the distraction stoped, no more cartilaginous tissue was present in the callus ,while the new bone tissues decline to mature.From scanning electron microscpy ,we observed the similar result. During the distraction period, the expression of basic fibroblast growth factor was observed in the osteoblasts on the newly formed one.The cells of osteoblastic and the mesenchymal cells in the distraction callus also expressed basic fibroblast growth factor.
