Objective:To explore the beta-fibrinogen gene -455G/A polymorphism and it's association with the susceptibility of cerebral infarction in young and middle-aged.Method: Patients were diagnosed according to diagnostic criteria. Genomic DNA in 78 subjects with cerebral infarction and 62 healthy controls were purified with DNA purification kits. Polymerase chain reaction of restricted fragment length polymorphism (PCR-RFLP) method was applied to determine the bi-allelic polymorphism in beta-fibrinogen gene -455G/A in patients and controls. The forward primer and reverse primer are 5'GAATTTGGGAATGCAATCTCTGCTACCT-3'and5'-CTCCTCATTGTCGTTGACACCTTGGGAC-3'PCR products were obtained with adequate PCR mixture and optimal PCR condition. The products were digested by restrictive enzyme and analyzed after electrophoresis.AA type got two bands 343bpand958bp; GA type got four band 343bp,383bp,575 and 958bp; GG type got three bands 343bp,383bp,and 575bp.The genotype distribution and allele frequency among different groups were compared..Results: The genotype (A/A, G/A and G/G)distribution ofβgene -455G/A polymorphism was0.04, 0.29and 0.67, in cerebral infarction patients, and0.03,0.24,and0.72 in controls(P>0.05). The allele frequency of allele in cerebral infarction patients was 0.19 and 0.15 in controls(P>0.05).Conclusion: The beta-fibrinogen gene-455G/A polymorphis was not significantly associated with the susceptibility of cerebral infarction patients.
