Chemical Study Of Hericium Erinaceur Oligosaccharide

Hericium erimaceus is a kind of medical plant.Accordingto the document,it is mainly cured intestinal canal diseasesuch as atrophic gastritis,duodenal ulcer,gastrelcomaetc.There have been a kind of medcine in the market which iscalled "Weilexin medicinal granules",is produced by theextractum of Hericium erimaceus.But a little research onchemical structure about Hericium erinaceur oligosaccharidewas reported up to now inside and abroad.The extractum of Hericium erimaceus was dissolved withwater.Then the supernatant was concentrated and precipitatedwith ethol,obtained precipitate (Hep) and supernatant (Hes).Hep was analysed by inorganic elements and ion chromatograph,the result show that there is trace SO42-.Hep was applied toDEAE-Sephadex A-50(HCO3-) column,and was fractioned intothree portions Hep-1,Hep-2 and Hep-3.Hep-1 had the mostsaccharide and yield,so it was taken more reseach.Hep-1 was applied to Sephadex G-25 column,and wasfractioned into three portionsHep-1-1,Hep-1-2 and Hep-1-3.The saccharide and yield of Hep-1-3 was the most,so it wasreseached.HPGPC showed that Hep-1-3 was a oligosaccharidemixture.Hep-1-3 was composed of Man and Glu.LC-MS showed that itwas made up from disaccharide to septsaccharide. Methlatedanalysis showed that it had (1→4,1→6)-linked as a mainchain,the branching point at 3-O.From the results of active experimentation,Hep is beteerthan Hericium erimaceus polysacchsride and positive medicinein curing chronic atrophic gastritis.Hep-1-3 was separated and purified through HPLC,thenobtained Hep-1-3-1(disaccharide) which had two peaks,andHep-1-3-2(trisaccharide) which had only one peak. Hep-1-3-1was composed of Man and Glu.GC-MS analysis showed that it had(1→4) and (1→6).So the structures of Hep-1-3-1 were Glu1→4Glu,and Glu1→6Man.Hep-1-3-2 was only composed of Glu,andhad (1→4,1→6)-linked,Glu was the terminal.EI-GC-MSanalysis showed that ,from the special ionic fragment peakof 305 of Hep-1-3-2,the structure of it can be determined,which was Glu1→4Glu1→6Glu.The seperation and purification of tetrasaccharide,pentasaccharide,hexasaccharide and septsaccharide,and thestudy of their structure is going on.
