As is well known, hepatitis B caused by HBV is very popular in the world. In Chinathe incidence of hepatitis B is extraordinary high, about 1.7 hundred million peoplewere infected with HBV, 170 million people are suffering from chronic hepatitis,and about 500 thousand people die of liver cancer or cirrhosis caused by HBVevery year. Immunication induced by inoculating HBV vaccines are veryeffective, and is one of the most successful practice in modern medicines byinterdicting chronic infection and reducing the incidence of acute HBV and deathof it.Well, among healthy people injected HBV vaccine, 14% of them showimmunological non-responsiveness or weak immune response with lessprotective antibodies producing. And doubt of HBV aberrance still exists. Itseems that it was in great needed to improve current HBV vaccineimmunogenicity or create a new kind of vaccine. According to relative research, HBV DNA vaccine, which can breakepeople's immunological tolerance and arise persistent humoral immunity and cellimmunity can be used for both prevention and therapy. It is a new point forfuture's research of HBV vaccine. So we create HBsAg subtype adr DNA vaccine, plasmid pVAX/HBS, usingwhich BALB/C mouse in test group were injected on week one, week two andweek four with reconstructed IL-II plasmid and recombinant IL-II protein asadjuvant with pVAX1 as control. The result showed that on week 4 HBsAbbecame appearance in mouse blood in test group but in control group it did not.In both test group and control group HBsAb test were minus.Our study believes that present HBV DNA vaccine can arise specific humoralimmunity on mouse, in which recombinant IL-II protein can enhance it to a highantibody level, but the effect of reconstructed plasmid of IL-II was weak.
