Tuberculosis(TB) has reemerged as one of the leading causes of death. Resistant Mycobacterium tuberculosis resulting from non-standardized treatment has become the most serious problem in anti-TB treatment. Exploring the molecular mechanism of resistance and the rapid detection methods now is the focus of tuberculosis control. Streptomycin(SM) and ethambutol(EMB) are important first-line anti-TB drug. It is well known that rpsL gene and embB gene are closely associated with the resistance of the Mycobacterium tuberculosis to streptomycin and ethambutol. The most common mutant site of rpsL gene in streptomycin resistant Mycobacterium tuberculosis is codon 43, while embB gene in ethambutol resistant Mycobacterium tuberculosis is codon 306. It is urgent to elucidate the variations in rpsL and embB in China as the genetic alterations vary with different countries and regions. Furthermore, detection of drug resistance by genotypes should be explored on the basis of local molecular epidemiology.
