Effects Of Recombinant Human Growth Hormone On The Growth Of Rectal Cancer In SRCA

Objective: To investgate the effect of recombinant human growth hormone(rhGH) alone and combined with 5-FU (5-flurouracil) on the transplanted tumor of human carcinoma of rectum. And study the effect of rhGH on the growth of human carcinoma of rectum in vivo and the feasibility of clinical application of rhGH in combination with 5-FU to patients suffering malignant tumor during the perioperative period.Methods: Forty five male mice BALB/C, which were subcutaneously injected CTX 100mg/kg per mouse before operation, were randomly divided into two big groups. The first big group thirty male mice BALB/C was randomly divided into five groups, INS group, IIrhGHl group, IIIrhGH2 group, IV rhGH1+5-FU group, V5-FU group. The second big group fifteen male mice BALB/C was also randomly divided into five groups in the same way. SRCA was carried out simultaneorsly with fresh explants of human carcinoma of rectum in all animals, at the same time the length and width of tumor explants in SRCA was measured in situ by anatomical microscope with ocular micrometer. The volume was calculated, using the standard formula (volume=length×width~2×1/2). From the first to the seventh day after surgery, rhGH1 group received daily i.h. rhGH injections (11U/kg), rhGH2 group received daily i.h. rhGH injections (3IU/kg), rhGHl+5-FU group received daily i.h. rhGH injections (HU/kg) and i.c. 5-FU (20mg/kg), and 5-FU group received daily .c. 5-FU (20mg/kg), while the NS group received daily i.h. and i.c. sterile saline injection at the same dosage. On the 10th day after surgery, the first big group mice were sacriced. The weight of all animals was measured every other day and the blood routines were also measured before animals executed.
