Ii blood-group system is the 27th red blood cell blood-group system. They do not have own code gene and are characterized as linear and branched repeats of N-acetyllactosamine, respectively. Therefore the i and I antigens are carbohydrate structures characterized. The newborn navel blood red cell carries i antigen. Conversion of the i to the I structure requires the activity of I-branchingβ-1,6-N-acetylglucosaminyl transferase (IGnT). The enzyme adds 2 N-acetyllactosamine (N-AC) to the linearity structure of i antigen, causing the andantol converting i active straight chains of carbohydrates to I-active branched chains.This research selects the navel blood red cell to make the immunity originally, carries on screening with the i (+) and i (-) red blood cell, obtains three anti- i monoclonal antibody. Through purifing i monoclonal antibody (IgG) to immunize the goat, we has obtained the anti-anti- i. The manufacture of anti- i monoclonal antibody and its two anti- have provided the necessary examination reagent for the examination rare blood-group ii.Successfully ,we have cloned IgnTc gene from the placenta through the RT-PCR method, and obtained pcDNA3.1-IgnTc. The reorganization material particle transfection the CHO cell, causes IgnTc in the CHO cell success expression. The IgnTc active appearance transformed the CHO cell surface i antigen into I antigen.
