Purpose To observe the effect of recombinant human bonemorphogenic protein-2 ( rhBMP-2) and Basic fibroblastic growth factor(bFGF) to repair full-thickness defects of articular cartilage defects in rabbits。Methods twenty-tour healthy white rabbits of 8-week-old were devided as randomly into 4 groups, Full-thickness defects of 4mm in diamete were created over the articular surface of both the femoral condoles by operation. After 4 weeks the joints was opened again and cleaning the fibrous tissues in the cavity. Fibrin glue(FG) was injected in the control group(A group),rhBMP-2/FG was injected into the knee cavity in B group,bFGF/FG was injected into the knee cavity in C group, rhBMP-2+bFGF/FGwas injected into the knee cavity in D group. All animals were killed after 4,8,12 weeks postoperatively after the injections,and the defects were examined grossly and microscopically. Studied systemically on the histology of cartilaginous regeneration.Results Result The repair results of defects were examined grossly and histologically after10,14and 18 weeks. In group A(control group) the defect was not repaired,and a dent on the surface ,no obvious new cartilage formation was seen, only a small quantity of new cartilage formation could be seen along the margin of defects . In group B the cartilage defects were smoothly repaired by white translucent tissue with obviously boundary between normal joint surface after 10,14 weeks, and the defects boundary were hard to identified after 18 weeks, of HE stain lots of mature hyline chondrocytes were found in extracellular matrix components( SEM) ,the histological feaftures of neocartilage were similar to that adjacent normal cartilage. In group C the cartilage defects were smoothly repaired by white translucent tissue, but it has one knee not entirely repaired when the rabbit was been killed after 10 weeks, it also had one which were observed after 14weeks ,the cartilage defects were smoothly repaired by white translucent tissue.
