Study On Correlation Between HCR Gene Polymorphism With Psoriasis Of Uigur In Xinjiang

Objective: In order to establish the foundation of researches on diagnosis,treatment and prognosis of psoriasis in gene level, the frequency of HCR allele in Uigur population from xinjiang area of China was realized;the susceptible gene of psoriasis in the 307 and 325 of HCR allele was sought,and the association of HCR gene exon 4 polymorphism with psoriasis susceptibility was analyzed.Methods: The difference between males and females on the age of first onset was analysed by using ofχ~2. One hundred and twenty-seven cases of psoriasis well-characterized patients with Ps and One hundred and eighteen healthy controls in xinjiang Chinese Uigur were studied. Polymorphisms of the DNA fragment of the HCR gene were detected by polymerase chain reaction (PCR) and the products were directly sequenced.The 307 and 325 C/T polymorphisms of all the cases were both detected by means of PCR-RFLP techniques.The direct count method was used for calculating the frequency of allele and genotype and Hardy-Weinberg test was made.Compared the genotype and allele frequencies between patients and controls .The significance of the difference between the genotypes and allele frequency was determined byχ~2.Results: The average age of the first onset was 27.08±13.49 for males, and 22.54±14.14 for females. Through the comparison of the gene polymorphisms between the 118 of psoriasis and 127 cases of controls,C-T polymorphisms at nucleotide 307 and 325 of the HCR region was identified. There were three genotypes, including CC, TT and CT. A significant different genotype distribution of 307TT and 325 TT polymorphisms were both observed between the two groups (P<0.05).Conclusions: The average age of first onset of PS was earlier in females than that in males.This study demonstrated that there was association between the polymorphisms of HCR gene exon 4 and psoriatic vulgaris in Uigur population of Xin jiang ,and the HCR allele...
