Purpose: To investigate the correlationship between DNA content detection of breast carcinomas by flow cytometry and histopathological parameter and hormone receptor, moreover,to analyse Clinicopathological significance of DNA content assay in breast carcinomas by flow cytometry. Methods: Collected the fresh tissue, paraffin embeded tissue and clinicopathological data of 65 cases breast carcinomas and 15 cases benign breast diseases in People's hospital of Xingjiang Uygur Autonomous Region from October of 2004 to January of 2006. Based on histopathological observation, DNA content were detected by using flow cytometry; at same time, estrogen receptor, progesterone receptor and ki-67 protein of breast carcinmonas were detected by immunohistochemical method.Results:(1)The proportion of DNA diploid of 65 cases breast carcinomas was 52.3%(34/65), which was lower than benign breast disease for the overall DNA diploid. In statistical analysis, the defference was significant (χ~2=11.069,P=0.001). (2) The mean S phase fraction of 65 cases breast carcinomas was 5.5887±4.3142, which was higher distinctly than benign breast disease, the difference was significant(t =2.997,P=0.001). The distribution of SPF was not same in different size of 65 cases breast carcinomas, the proportion of high SPF beyond 20mm was higher, the difference was significant (χ~2=4.584,p=0.032);In histological gradeⅠ~Ⅱ, high SPF was few, in histological gradeⅢ,high SPF was more, the difference was significant(χ~2=9.256,p=0.002); In different age, lymph node status and different expression of hormone receptor, the proportion of high SPF is not same, but absent statistical significance. (3)DNA ploidy distribution was not same in different type breast carcinomas ,all the special type breast carcinomas were diploidy ,but the difference was no significance; There were no statiatical significance between DNA ploidy and clinicopathological parameter, hormone receptor in spite of dissimilitude distribution of DNA ploidy. (4) The expression of ki-67 protein by immunhistochemical method was obviously different in groups of different size and histological grade(χ~2=4.623,9.158;p=0.032,0.002).Conclusion:(1) The appearance of DNA aneuploid of breast tumors may be used as a reference when estimated malignant breast tumors, and it would help judge the early lesion.(2) Through the correlation with clinicopathological parameter, proliferation index SPF was more...
