Objective: to explore the relationship among eIF-4E gene and VEGF-C express, angiogenesis, lymphangiogenesis and lymphatic metastasis in breast cancer.Methods: Western-blot and immunohistochemical method was used to detect the expressions of eIF-4E, VEGF-C and the density of micro vessels and micro lymphatic vessels in 57 specimen of breast cancer. the eIF-4E ,VEGF-C expression and the MVD ,LMVD were quantified.Results: Western blot results indicated that both lymph node positive and negative group had 25 KD-eIF-4E protein strips; the O.D value of positive group is significantly higher than that of negative group (10.11±0.30 and 5.94±0.23, t=5.37,P<0.01). In the positive group, the VEGF-C positive cancer cell distributed extensively and was stained deeply (4.20±0.98). MVD value in lymph node positive group was 32.28±7.41, significantly increased comparing with that of negative group (24.67±8.12, t=2.51,P<0.05). LMVD in lymph node positive group was notably higher than that in the negative group (25.28±5.33,17.40±4.87,t=2.41,P<0.05). Correlation analysis indicated, the level of eIF-4E is positive correlated with VEGF-C expression, MVD, LMVD and lymphatic metastasis of breast cancer (B= 0.635,0.325,0.760 and 0.570 respectively, P<0.05).Conclusions: eIF-4E gene may induce the angiogenesis and lymphangiogenesis by increasing the expression of VEGF-C, by which regulating the lymphatic metastasis of breast cancer. Blocking the transmission of VEGF-C/VEGF-2/VEGF-3 signals may become the effective way of inhibiting the lymphatic metastasis of breast cancer.
