Objective To investigate the expression of Ets-1 and MMP-9 in gastric cancer and their relationship with tumor invasion and metastasis, and using antisense oligodeoxynucleotide technique to explore the effects of Ets-1 ASODN on the human gastric cancer cell line BGC-823 by analyzed the expression changes of Ets-1 mRNA and Ets-1\MMP-9 protein. Through this research, We hope to explore the possible role of Ets-1 in the invasion and metastasis of gastric cancer and its mechanism. Methods Part 1: Immunohistochemical staining (S-P method) for Ets-1 and MMP-9 were performed in 97 cases of GC, adjacent tissues and 28 cases of normal mucosa. Part 2: A 20mer ASODN, targeted to the specific sequence of Ets-1 mRNA, was transfected into human gastric cancer cell line BGC-823 using lipofectmine 2000 transfection reagent, then the changes of Ets-1 level both in mRNA and in protein, MMP-9 level in protein were analyzed respectively. Results Part 1: (1) The positive rate of Ets-1 and MMP-9 immunostaining in gastric carcinoma were higher than that in adjacent tissues and normal tissues(P<0.01). (2) There was a difference of the expression rate of Ets-1 and MMP-9 among papillary adenocarcinoma , tubular adenocarcinoma , poor differentiated adenocarcinoma and mucoid carcinoma (P<0.01) . (3)The positive expression rates of Ets-1 and MMP-9 were significantly related with the depth of tumor invasion, lymph node metastasis and clinical stage (P<0.01), but they did not correlate with sex, age, tumor size and distant metastasis (P>0. 05) .
