Objective: To observe the injury of deformability, membrane fluidity, membrane protein expression, membrane ATPase and the concentration of Na+ and Ca2+ in rat erythrocyte which were caused by acute hypoxia and the protective effect of hypoxia acclimatization on them.Methods: Healthy male Wistar rats were randomly divided into normal group, acute hypoxia group and hypoxia acclimatization group, each ten ones. They were respectively exposed to hypoxia and trained to acclimate to hypoxia in the hypoxia-room simulating high altitude and blooded from their hearts after anaesthetized. Haematology parameters were measured by F-820 automatic blood analysis apparatus and erythrocyte deformability was measured by LG-B-190 blood cell deformability instrument. DPH which showed membrane fluidity was detectd by F-4000 fluorometry. Cholesterin and total lipid were detected by U-2001 ultraviolet spectrometer. Lipid components of erythrocyte membrane were measured by TLC, which included phosphatidylcholine (PC), phosphatidylethanolamines (PE), phosphatidylserines (PS) and sphingomyelin (SM). The two-dimensional electrophoresis maps of the rats erythrocyte membrane protein were achieved. The different protein spots were found by Image Master 2D Elite and identified by mass spectrum. Erythrocyte membrane ATPase and the concentrations of Na+ and Ca2+ in it were measured by relevant reagent kits respectively.Results: 1. In acute hypoxia group, the total sum of erythrocyte, hematocrit and hemoglobin increased; the deformability, membrane fluidity, the content of membrane cholesterin and total lipid and the ratio of cholesterin and total lipid (C/P) declined, the content of all the components in erythrocyte membrane lipids were altered.
