Objective: To explore the therapeutic mechanism of Naloxone on rats suffered in ARDS caused by oleic acid,we observed the change of lung W/D and the chang in morphopathology, detected TNF-α,Il-1β&ET-1 in serum,the content of MDA in lung.Method: 30 healthy Wistar rats(weighed 200-250g) were divided into three groups randomly(n=10).1.Control group:Anesthetize rats with urethane(1.0g/kg) through abdomen.Separate one side femoral vein and artery.And then inject normal saline(NS4.1ml/kg) through femoral vein.2.Oleic Acid(OA) group:After anesthesia and disconnection like above,inject oleic acid(0.1ml/kg) and NS(4.0ml/kg).3.Naloxone group:After anesthesia and disconnection like above,inject oleicacid(0.1ml/kg) and naloxone(lmg/kg,diluted in 2.0ml/kg of NS).Then three hours later draw blood through abdominal aorta to obtain serum for TNF-α ,IL-1β &ET-1.Take out all lung to observe the chang of shape.Then weigh, dry and weigh again the left lung.cut right lung for optics microscope and transmission electric microscope,and for detecting MDA.Result:1.OA and naloxone group showed the left lung W/D,the content of TNF-α ,IL-1β &ET-1 in blood and the content of MDA in lung higher than the control group,The differents between OA&naloxone groups and control group was quite significantly(p<0.01).Between OA and naloxone group,the different was quite significantly too( p<0.01 or 0.05).TNF-α &H-1β in serum and MDA in lung correlate to the left lung W/D(p<0.01 or 0.05).MDA in lung also correlated to the content of...
