Abjective:Discuss the changes of serum IL-6, TNF-α, NO, iNOS and lung tissues of SOD and MDA content on rat ARDS.and through the application of baicalin after oleic acid induced ARDS rats model, the variation of the above indicators further defined baicalin the influence of the experiment.Methods:30 healthy male SD rats were randomly divided into model group, normal group and baicalin group. By the method of anaesthetized normal group rats with injected 1.0 g/kg normal saline in left lower quadrant abdominal.and spin out the femoral vein catheter step by step, after then saline injection of 0.1 ml/kg,8.75 ml/kg and physiological saline injection the abdominal cavity. With the same anesthesia, stripping femoral vein and femoral vein catheter method dealing with model group, then 0.1 ml/kg of oleic acid injection in femoral vein, ARDS model is set up, and finally in the abdominal cavity injection of 8.75 ml/kg of saline solution. Baicalin group in the same way to produce oleic acid ARDS rats model, the abdominal cavity injection of 350 mg/kg of baicalin.Three hours after the end of the experiment, the blood is drawn by the abdominal aorta, extract serum, save medical refrigerator of 40℃ below zero. After taking blood to be put to death animals.Determination of rat lung wet to dry weight ratio after removed after thoracotomy. Another part of lung tissue storage at -65℃ in a freezer, a small number of lung tissue were soaked in 10% acetic acid uranium and formaldehyde solution in order to make specimens of electron microscopy and light microscopy.ELISA method were used to detect serum TNF alpha and IL-6, colorimetry method were used to detect serum NO and iNOS, SOD content in lung tissue WST-1 method, with sulfur content of MDA in lung tissue generation barbituric acid method.Results:1. Compared with normal group, lung wet/dry weight ratio of model group increased significantly (p< 0.01),the baicalin group of lung wet/dry weight ratio difference was statistically significant (p< 0.05), the contents of serum TNF-a model has significant difference (p< 0.01), the contents of serum TNF-a of baicalin group has not statistically significant difference (p> 0.05), MDA content in lung tissue and serum IL-6 content, NO, iNOS were significantly increased (p< 0.01), SOD content in lung tissue decreased obviously (p<0.01).All indicators baicalin group compared with oleic acid group were the significant differences (p<0.01).2. Observe the general form of the lung tissue pathology:the normal group lung tissue were pink,. The lung volume of model group were obviously increased,and edema and the lung tissue bleeder and edema.The faces of lung tissue be seen visible flake bleeding, section has a light yellow or reddish liquid overflow;Baicalin group compared with model group significantly reduce the lesions. Light microscopy:the lung tissue structure of normal group were clearly and the complete alveolar interval no congestion, edema, inflammatory cells infiltration, in model group that the lung tissue structure disorder small bronchi and pulmonary edema around small blood vessels, and numerous inflammatory cells infiltration;Alveolar interval obvious thickening, swelling, and pulmonary interstitial also showed thickening, edema, inflammatory cells infiltration, alveolar capillary hyperemia expansion, The above pathological changes, baicalin group significantly reduce d than model group.Electron microscopy (sem):Model group the II alveolar cell damage seriously,and a lot of obviously phenomenon of vacuoles and visible emptying.The above performance in baicalin group were lighter in the model group.Conclusion:The serum of TNF-α, IL-6, NO and iNOS and the lung tissue of MDA and SOD are reaction degree of lung tissue damage in ARDS.Baicalin by exerting its anti-inflammatory, antioxidant effect, reduce the NO content in serum of rats, iNOS activity, thus reducing the inflammatory cytokines release, increase SOD activity in the lung tissue, reduce MDA content, eventually played a protective effect on acute respiratory distress syndrome...
