Abjective:The observed of TNF-α, IL-1β, IL-6, SOD, MDA content in TMP oleic acid induced acute respiratory distress syndrome.Methods:The number of30adult male SD rats were randomly divided into normal group, model group and TMP group,there were10rats in each group,model group intravenous injection tube oleic O.lml/kg, made a rat model of ARDS. TMP group of rats injected intravenously tube oleic acid injection TMP injection20mg/kg. Normal rat femoral vein injection of saline3.1ml/kg, three hours later extracting the blood serum collected in the abdominal aorta, the-40℃refrigerator. Take blood and the animals were killed, after thoracotomy removal of lung tissue wet weight,72hours later weigh as the lung dry weight. Said the latter part of the lung tissue wet weight placed-65℃refrigerator, part of the lung tissue placed in10%neutral formalin and uranyl acetate to prepare light microscopy and electron microscopy specimens. Serum by ELISA serum TNF-α, IL-1β, IL-6levels, the use of SOD in the WST-1assay in lung tissue content, using the thiobarbituric acid assay content of lung tissue MDA.Results:1.Compared with the normal group, TMP, model lung wet/dry ratio was significantly increased serum TNF-α, IL-1β,MDA content in lung tissue IL-6levels were significantly increased; lung tissue SOD activity was significantly reduced, there are significant differences; these indicators were significant differences(P<0.01).2.Lung histopathology observed Broadly:Normal lung tissue was pink, surface finishing, no bleeding and edema, model group significantly increased the volume of lung tissue, dark red, flaky visible bleeding, cut yellow or pale red liquid spill; TMP group compared with the model group of these lesions was reduced. Light microscopy:Normal lung tissue structural integrity, alveolar septa no congestion, edema, no inflammatory cell infiltration in lung tissue model structural disorder, pulmonary edema and small bronchi and perivascular inflammatory cell infiltration; edema thickening of the alveolar septa pulmonary interstitial thickening, edema, inflammatory cell infiltration, alveolar capillary dilation, congestion; cavity protein edema fluid and neutrophil exudation, hyaline membrane, the pathological changes of light TMP group compared with the model group. SEM:Normal alveolar type Ⅱ cells and endothelial cells without necrosis, edema phenomenon. Model group II alveolar cell damage seen a lot of vacuoles, a small amount of emptying phenomenon can be seen clearly. TMP group compared with the model group mitigate these changes.Conclusion:TMP on rat ARDS more effective treatment, the mechanism by inhibiting TNF-α, IL-1β, IL-6inflammatory cytokines, reducing the activity of MDA in lung tissue, increased SOD levels, which play an anti-inflammatory and antioxidant effect.
