Objective To investigate the surviving area and its mechanism, the microcirculation dynamic changes and ultrastructural changes of expanded prefabricated flaps. Methods A total of 36 New Zealand male rabbits were divided randomly into expanded prefabricated (group A), expender lined (group B) and prefabricated (group C) groups, each group consisting of 12 rabbits. After the femoral vascular bundles were transplanted into subcutaneous tissues of abdomen, expanders were implanted into the deeper dartos. For the expanded prefabricated group, the expansion was carried out at 7 days postoperatively. After 52 days, when the expander was fully expanded, island flaps with the prefabricated vessels as the pedicles were formed. The flaps were measured by digital recording of survival area, laser Doppler flowmetry (LDF) , immunchistochemistry , light microscope and transmission electron microscope (TEM).
