Inhibition Of Tongyou Decoction And Its Separated Formulas On EGFR-PLC-γ1 Signalling In Esophageal Carcinoma Cells EC9706

Objective: By observing inhibiting effects of Tongyou decoction and its separated formulas(promoting blood and Qi circulation and nourishing yin and blood) on the cell proliferation of human esophageal cancer cell EC9706 and EGFR-PLC- Y 1 signalling in this cells, the molecular mechanism of Tongyou decoction treating esophageal cancer was studied to understand esophageal change in traditional Chinese medicine.Methods: After esophageal cancer cell line EC9706 cells are treated with Tongyou decoction and its separated formulas comparing with control group, the morphological changes of the cells are obverted under a convert microscope, the cells growth are examined by MTT assay, the protein expression EGFR, PLC-γ1, PKCa and protein phosphorylation level are analyzed by western blot method.Results: 1. The EC9706 cell proliferation were significantly inhibited with the decoction and its promoting blood and Qi circulation with smaller and shinrinkage shape, larger space among the cells, more cell debris, smaller cell clone and converging rate comparing with the control group, in which the effect of the promoting blood and Qi prescription is stronger than the whole formula in dose and time dependent. The maximum inhibition rates of the decoction and its promoting blood and Qi circulation are71.5% and 81% respectively. The effect of nourishing yin and blood prescriptions on the cell growth is weak at maximum dosage (3200μg/ml)with similar shape to the control group. These results show promoting blood and Qi circulation medicine play main role the growth inhibition of Tonyou decoction on esopheageal caner cells.2. high expressions of EGFR, PLC-γ1 and PKCa are found in the esophageal cancer cell line EC9706 cells.
