[Objective]To investigate the effect of Topiramate(TPM) on bone development in growing rats, which may contribute to monitoring the adverse effects of TPM.[Methods]60 3-week-old male Wistar rats were weight matched and randomly divided into 4 groups. The rats in the study groups were administered intragastrically with TPM 40mg/(kg·d) and 80 mg/(kg·d) respectively, while the rats in the positive group with Phenytoin 80 mg/(kg·d) and the ones in the negative group with distilled water partes aequales. After 3-month administration the changes of bone metabolism were assessed using indices as follow: biochemical markers in serum,bone mineral density and biomechanics.[Results]1. Effect on body weight: Body weight gain of rats taking TPM was lower than that of rats in negative control group (P<0.05),and rats with high dose of TPM were affected more significantly than ones with low dose (P<0.05).2.Effect on biochemical markers of bone metabolism in serum: The levels of bone alkaline phosphatase(BALP),osteocalcin(OC),tartratercsistant acid phosphatase(TRACP) and calcium in serum of rats in study group were higher than those of rats in negative group(P<0.01), while High dosage of TPM group had been influenced more than low... |
