Objective To study the expression of Livin in gastric carcinoma, and investigate it's role tumorigenesis, progression in gastric carcinoma and it's mechanism of resistance to chemotherapy,to supply a potent new target for therapy for gastric cancer. immunocytochemistyMethod (1) Detected the expression of Livin in SGC-7901 cells by immunocytochemisty(ICC). (2) Livin mRNA and protein expression in 30 cases of gastric cancers and sideward tissues of cancers and 6 cases these benign gastric tissues were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and western-blot. (3) Proliferation of SGC-7901 cells treated with 5-Fu was assessed by methyl thiazolyl tetrazolium (MTT) colorimetry. (4) The apoptotic cells and distribution of cell cycle were detected by flow cytometry. (5) Livin mRNA and protein expression in SGC-7901 cells before and after treated by 5-Fu were detected by RT-PCR and western-blot.Result Livin protein was expressed in nucleolus and cytokinesis of SGC-7901 cells.The positive percent of Livin expression was 66.67%(20 / 30 cases),which had significant difference (P<0.05).
