Radix Angelica dahurica is one of the commonly used traditional Chinese medicines. It has analgesic and antiinflammatory effects. Imperatorin is a biologically active furanocoumarin from the roots of Angelica dahurica. In this paper, a method of quality assessment of Radix Angelica dahurica was developed, as well as the pharmacokinetics of imperatorin was studied in rats.1. Studies on the quality assessment of Radix Angelica dahuricaA method for simultaneous determination of imperatorin and isoimperatorin in Radix Angelica dahurica by HPLC was used. A ULTRON VX-ODS column (4.6 mm×250 mm, 5μm) was used with the mobile phase consisting of acetonitrile-water (65:35, v/v). The flow rate was 0.8 mL·min-1, and the detection wavelength was set at 247 nm. The linear ranges of imperatorin and isoimperatorin were 5-50μg·mL-1 (correlation coefficient r = 0.999 8) and 2-20 μg·mL-1 (correlation coefficient r = 0.999 9), respectively. The average recoveries of imperatorin and isoimperatorin were 98.4 % ( RSD =1.2 %) and 96.3 % (RSD = 1.7 % ), respectively. The determination of the contents of two coumarins provide a quantitative basis for the quality assessment of Radix Angelica dahurica...
