19 Ligusticum chuanxiong Hort. samples of different specifications were collected from different habitats and the quality assessment was developed.The conditions for the extraction of tetramethylpyrazine present in the extract were studied. The Water refluence is more optimal than Ultrasonic extraction. The reflluence extraction method was further studied by orthogonal design. Four factors were selected in this experiment, including the water and alcohol of different concentration, the solvent consumption, time of extraction and times of extraction. The result showed that the optimum extraction condition was 80% alcohol consummated six times the amount of material, and two times for tow hours each time. The extraction conditions of Ferulic acid and vanillin using ASE (accelerate solvent exactor) were studied. Three factors were selected in this experiment, including the static extraction time, cycles and temperature. The result showed that the optimum extraction condition was 80% alcohol, and two cycles for 15min's static extraction time at temperature of 100℃ each time.Tetramethylpyrazine, Ferulic acid and Vanillin in Ligusticum chuanxiong Hort. were determined by RP-HPLC. The linear range of Tetramethylpyrazine was 0.3-24μg·mL-1(r=0.9995). The average recovery (n=9)was 97.5% (RSD = 1.8%). The result showed the calibration curves of Ferulic acid and Vanillin were linear in the range of 2.8-168μg·mL-1 and 0.1-6μg·mL-1 respectively.
