Chuanxiong is the dry rhizome of Ligusticum chuanxiong Hort.,which is used for the treatment of chest pain,heartache,headache,rheumatic pain,irregular menstruation,etc.It was Commonly used as traditional Chinese medicine.At present,the planting area of Ligusticum chuanxiong Hort.in the whole province has reached more than 100,000 mu,and the annual output is about 20,000 tons.It has the highest output in all the raw materials of Sichuan,the second output value.In the process of storage,Ligusticum chuanxiong Hort.is easy to occurrence of insects,mildew and other deterioration,which seriously affects the effectiveness and safety of clinical medication.In this study,based on the natural conditions,and different temperature(15 ?,25 ?and 35 ?)and different humidity(40%,60%,80%),study the change of chemical composition and the characteristics and isothermal adsorption curve of,in order to establish early warning model in the process of Ligusticum chuanxiong Hort.And by identified the surface of the mold and moth species on the storage process,wish to Provide scientific evidence.The research contents are as follows:1 Changes in appearance traits during storage of Ligusticum chuanxiong Hort.The traditional eye,hand touch and nose smell method were used to identify the texture,section color,odor,insect mites,mildew and oil in the storage process.The research shows that under natural conditions,After the two mouth(August),insects have been occurred.Under different storage conditions,when the temperature is more than 25 ?,humidity is more than 60%,the mildew,oil phenomenon have been occured,its mildew phenomenon mainly appear in the humidity of 80%.When temperature is less than 25 ?,humidity is less than 60%,Ligusticum chuanxiong Hort.imedicinal have no obvious deterioration phenomenon.2 Changes of chemical composition of Ligusticum chuanxiong Hort.during storage2.1 Changes of total fluorescence substancesThe cross section of Ligusticum chuanxiong Hort.shows lilac fluorescenc e under ultraviolet light,which is an important characteristic of traditional phys icochemical identification of Ligusticum chuanxiong Hort..This research adopt s the fluorescence spectrophotometer measured samples of Ligusticum chuanxiong Hort.total fluorescence material content in the process of storage,research has shown that under natural conditions,along with the storage time increased as well as the bug eat by moth phenomenon obviously,decrease its total fluor escent substances content in Ligusticum chuanxiong Hort.,bug eat by moth before its content range of 79.07% ~ 48.55%,bug eat by moth after its content range of 51.63% ~ 35.42%;Under different storage conditions,when the temp erature is more than 25 ?,when humidity is more than 60%,with the increase of temperature and humidity,Ligusticum chuanxiong Hort.total fluorescent substances content in a downward trend.2.2 Chemical composition changes of alkaloids,phenolic acids and phthalidesIn this study,HPLC method was used to determine the content changes of ligustrazine,phenolic acids(chlorogenic acid,ferulic acid,ferulic acid conifer ous)and phthalide(z-ligustilide,ligustilide I,H,A,and n-butylphthalide and butylphthalide)in ligustrong during storage.The results showed that under natur al conditions,the butylphenthalide,z-ligustilide,ligustractone A,ligustractone I,ligustractone H and organic phenolic acid compounds in ligusticum chuanxion g showed different degrees of decreasing trend,but ligustrazine showed A certain rising trend,and there was no significant change in n-butylphthalide.Under different storage conditions,the contents of ligustrazine,ligustractone I and lig ustractone H in chuanxiong showed a certain increasing trend during the storag e process,while the contents of chlorogenic acid,ferulic acid,z-ligustilide,ferulic acid pine and cyperus and butylphenylphthalide all showed a decreasing trend.The content of butylphthalide did not change obviously.3.Establishment of early-warning model for Ligusticum chuanxiong Hort storage3.1 Discriminant analysis and linear regression modelIn this study,discriminant analysis(DFA)was used to discriminate and classify different samples of Ligusticum chuanxiong Hort.with varying degrees of metamorphism,such as chlorogenic acid,ferulic acid,coniferyl ferulate,yogh urtlactone I,yoghurt lactone H,and yoghurt Ester A and total fluorescent sub stances are metamorphic indicators.The study found that the established DFA discriminant model has a correct rate of 96% for unknown samples,and can distinguish between different metamorphic samples of Ligusticum chuanxiong Hort.The linear regression equations of ferulic acid,sedative lactone I and Z-dec alactone with storage time were established,which can predict the ferulic acid,sedative lactone I and Z-ligustilide during storage of Ligusticum chuanxiong.The content,and the sample "slight metamorphism" as the early warning zone,set the ferulic acid content to 0.1%,and the Z-decanolide content to be 1.77% as the warning value.3.2 Study on the isotherm adsorption curveLigusticum chuanxiong Hort.is also prone to mildew during storage.Microbial reproduction is closely related to water activity.In this study,the water activity of the Ligusticum chuanxiong Hort.sample with different initial water content was 0.11~0.90,and the ambient temperature was 25 ?.The isothermal adsorption curves at 35 ? and 45 ? were used to investigate the relationship between the equilibrium moisture content and water activity of different initial water contents at different temperatures,and predict the safe moisture content.The results show that the equilibrium moisture content of the Ligusticum chuanxiong Hort.sample increases with the increase of water activity at different temperatures,the isothermal adsorption curve belongs to the “III” type,and the Ferro-Fontan model adsorbs the different initial moisture content.The isotherm al curve fits optimally.4 Identification of mold and aphid species on the surface of Ligusticum chuanxiong Hort.In this study,for the first time,the molds and insect pests were identified by the molecular biology of the molds and insects.The results showed that there were 2 strains of mold and aphids on the surface of Ligusticum chuanxiong Hort.during storage,which were fungi of Basidiomycotina.Trametes hirsute,Aspergillus oryzae,and Silvanus bidentatus of the genus Spirulina.
