In the present dissertation, the anticancer constituents of ALN, a Chinese medicinal herb, were examined for their cell-cycle inhibitory and apoptosis-inducing activities using K562 cells, a human chronic myelgenous leukemia cells.Chinese herb ALN belongs to Legume family. It was reported that the chemical constituents from the bark of the same genus of plants possess antitumor activity. So far there was no report about the components of ALN and its bioactivities. Then this study was designed to originally systematic investigate the active constituents of ALN on human tumor cells K.562. In the process of isolation, guided by bioassay of cell-cycle inhibitory and apoptosis-inducing activities using K562 cells, we determined the active parts and fractions then confirmed the isolating scheme for active components by micro-quantity test. Nine compounds were isolated and their chemical structures were respectively identified on the basis of the physico-chemical properties and spectral data as: (-)Budmunchiamine K (1), (+)9-Normethyl budmunchiamine K (2), Phthalic acid dibutyl ester(3), Phthalic acid bis-(2-estyl-hexyl) ester(4), 3-O-β-D-xylopyranosyl-(1→2)-β-D-fucopyranosyl -(1→6)-β-D-2-aceto-glucopyranosyl echinocystic acid (5), 21-O-( 2,6-dimethyl -8-methoxy-2,6-octadienoyl)-acacic acid (6), 21-O-(2,6-dimethyl-8-methoxy- 2, 6-octadienoyl)-3 -O-α-D-2-aceto-glucopyranosyl-acacic acid(7), 4'-Hydroxy-tr ans-cinnamic acid octacosyl ester(8), Stigmasta-7,22-dien-3-one(9). Compounds 1-9 were isolated from ALN for the first fime. In these compounds, 2,5,6,7 were new compounds, and 3, 4, 8, 9 were firstly isolated from the same genus of ALN.On the basis of previous chemical study, inhibitory effects of compounds on the proliferation of human cancer cells were evaluated by SRB method and their effects on cell-cycle and apoptosis were investigated by flow cytometry. The anticancer activities of the nine compounds on K562 cells were examined and their minimum inhibitory concentrations were assayed with the multiple...
