Eczema is a common skin disease , the etiological factors of eczema have been entirely clear, it is caused by chemical irritation and allergic action, and it is believed to be related with immune defect, infection and heredity. Monoammonium glycyrrhizinate(MAG) is one of active component in Glycyrrhiza uralensis Fischer. The glucoside with hormone activity in its molecular structure is similar to the basic mother nucleus of adrenalin and has the functions of anti-bacterial, anti-inflammatory action, anti-virus, anti-allergic action and stopping the itching, which are closely related to the recovery mechanism of eczema. We studied the formulaton and technics about Monoammonium glycyrrhizinate(MAG) gel and made the primary study on the therapy mechanism of eczema with monoammonium glycyrrhizinate.In the paper, the reaction between monoammonium glycyrrhizinate and Carbopol were determined by Differential Scanning Calorimeter. The assay method of monoammonium glycyrrhizinate gel and the release of MAG from gel was established. We studied the influence of Carbopol, propylene glycol and absolute alcohol on MAG gel in the formulation, So the formulation of monoammonium glycyrrhizinate gel was optimized by orthogonal design experiment.On the basis of optimized formulation above, the appearance, pH, viscosity and content of monoammonium glycyrrhizinate gel was determined, and the release of MAG from gel as well as the stability of MAG gel was studied. The results showed that the basic characteristics of MAG gel, the drug content and the release of MAG had no obvious changes in high temperature experiment, accelerated experiment and in long-term experiment at 25±2℃ for 6 months, but the strong light can reduce the viscosity of MAG gel, and the release of MAG from gel in vitro was up to Higuchi equation.The results of allergic and irritation experiment demonstrated that MAG gel had no allergic action and irritation to intact and damaged guinea pig skin, and it is safe for clinical application.We have evaluated a subchronic model of contact hypersensitivity in the guinea pig to mimic human chronic/recurrent eczema.
