ObjectiveTo investigate the association between the 759 T→G→C , 1078A→G, 1233A→G simple nuclic polymorismphs(SNPs) of angipoitin-2 with type 2 diabetes mellitus (T2DM)and diabetic nephropathy (DN) in Kunming Han nationality. MethodsSurveyed SNPs of the angiopoitin-2 gene in 221 cases of Kunming Han people with type2 diabetes mellitus , met the criteria of the World Health Orignization (WHO) of 1999 and control cohort consisting of 104 cases. (1) Genotype and allelic frequency of angiopoitin-2 in extron 2 site in 759, extron 4 site in 1078, extron 5 site in 1233 were determined in 221 in type 2 diabetic patients including 134 diabetic nephropathy and 104 heathy subjects by PCR-ARMS (amplification refractory mutation system PCR)and PCR-RFLP(polymerase chain reaction restriction fragment-lenth polymorphism). (2) the blood glucose insulin c-peptic at 0 min, 30 min,60 min and 120 min, respectively. Blood pressure, TG, TC, HDL-C, LDL-C, HbAlc, body mass index and waist heap rate were examined. (3) Serum level of IL-6 and angiopoietin-2 were measured by ELISA(enyme linkage immure sorble assary). (4) homeostatic model assessment for insulin resistance (HOMA-IR)were also determinedResults(1)The C and G allele frequency of type 2 diabetes and diabetic nephropathy patientswere significantly higher than that in healthy subjects in 759 T→G→C.(2)There were no difference of the AG + GG genotype, G allele frequency (NC vs DN...
