Objective The purpose of this project is to find new STR locus useful and applicable in Forensic Genetics, to establish the fluorescent Multiplex STR system of D12S1064, GATA140E03 and D18S536. According to the guidelines of TMGDAM in United States, we also validate the forensic application of this multiplex system. Methods First, we undertake the population genetic research of five STR loci, D9S1119, D12S1064, D18S536, GATA156D11, andGATA140E03. Second, we select three loci, D12S1064, GATA140E03 and D18S536, with good polymorphisms and high Discriminate Probability, and begin the research of this fluorescent multiplex system using the traditional multiplex methods. Last, we detect the sensitivity, accuracy, species specificity, and different PCR outcome under each anneal temperature.
