Objective The purpose of this project is to find new STR locus which are useful and applicable in Forensic genetics, and to establish the multiplex STR system of DYS717 and DYS726. Based on the guidelines of TMGDAM in United States,we validate the forensic application of this multiplex system.Methods At the first step , we undertook the population genetic research of five STR loci, DYS711,DYS712, DYS714, DYS717 and DYS726. And then, two loci, DYS717 and DYS726, with good polymorphisms and high discriminate probability were analyzed by traditional multiplex methods in which two loci were amplified in one tube and the products were separated by PAGE gel.Results We obtain the genetic parameter such as gene frequency, Discrimination Probability, Probability of Exclusion, which are very useful in forensic casework. We established the multiplexsystem of DYS717 and DYS726. Moreover, it is easy to get the PCR products in two loci multiplex system by domesitc Taq polymerase without the hot start. Therefore, we provide a much cheaper multiplex PCR system with high amplification efficiency.Conclusions This multiplex system can get the reliable genetic data. The forensic genetics research implies that it is an easily-regulated, low cost multiplex system with stable outcome, high sensitivity and accuracy, good species specificity. Based on the above advantages, it could be recommended to the forensic genetics practice.
