Objective The purpose of this project is to do concordancestudies between miniSTR and common STR of THO1,TPOX,CSF1PO loci,to study the fluorescent multiplex system of mini-TUO1, mini-TPOX,mini-CSF1PO loci, to validate the forensic application of thismultiplex system. Methods The primer design is referring to thearticleof Bulter. Over 120 samples provided by the laboratoryare detected to verify the concordant results between miniSTR andcommon STR. We construct the miniSTR multiplex system usingdomestic Taq DNA polymerase, and detect the results by ABI—310Genetic Analyzer. We have alsostudied the sensitivity, accuracy, species-specificity and different PCR outcome under each annealtemperature. Results Over 120 samples have verified that thethree miniSTR can provide full concordant results to commercialSTR kit. We have successfully constructed the fluorescentmultiplex system of mini—THO1 mini-TPOX and mini-CSF1PO. It showsthat the anneal temperature is wide (60℃is the best), thesensitivity is 0.25ng template DNA, the species-specificity isgood, and it can be used to detect aged bloodstain sample.Conclusions The results between miniSTR and common STR of THO1,TPOX, CSF1PO Loci are concordant. The miniSTR multiplex system canbe detecded by ABI—310 Genetic Analyzer, having high sensitivity,stable results and good species-specificity. Generally, thisminiSTR multiplex system can be recommended to the practice offorensic genetics.
