Objective: To investigate the expression of proliferating cell nuclear antigen (PCNA) and transforming growth factor-betal (TGF-β1) in human gastric carcinoma and its clinical significance.Methods: The expression of PCNA and TGF-β1 protein in 60 gastric carcinoma tissues and 20 adjacent normal gastric mucosal was examined by immunohisto-chemical method. Statistical analysis of group differences was performed by x2 test.Results: Compared with the adjacent normal gastric mucosal tissues, the expression of PCNA and TGF-β1 in gastric carcinoma was significantly higher(P< 0.01). The positive rate of PCNA in gastric carcinoma was 71.7%, and that of TGF-β1 was 65.0%. However the positive rate of PCNA in adjacent normal gastric mucosal was 20%, and that of TGF-β1 was 15%. PCNA and TGF-β1 expression was associated with the stage of TNM, histological differentiation, invasion depth and lymphatic metastasis of gastric carcinoma(P<0.05).In patients with positive PCNA expression, the 1, 3, 5 years survival rates were significantly lower than that in patients with negative PCNA expression(P<0.05). In patients with positive TGF-β1 expression, the 1, 3, 5 years survival rates were also significantly lower than that in patients with negative TGF-β1 expression(P<0.05).Furthermore, the expression of PCNA was positively correlated with the expression of TGF-β1 In patients with positive PCNA and positive TGF-β1 expression, their malignancy grade and metastasis potential were significantly higher.Conclusion: The high expression of PCNA and TGF-β1 is related to the malignant transformation and proliferation in gastric carcinoma. They may be useful to evaluate the biological behaviors and predict the prognosis of gastric carcinoma.
