Objective: to study the expression of proto-oncogene Bmi- 1 and proliferating cell nuclear antigen (PCNA) in normal cervix, cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma, and analyse the clinical significance and relationship of proto-oncogene Bmi-1 and PCNA in the process of the cervical lesions and as well as in cervical squamous cell carcinoma.Methods:collect 96 cases of paraffin embedded cervical tissue specimens which were obtained from cervical cancer surgery and cervical biopsy during 2011-2013 at the department of pathology in the First Hospital of Lanzhou University and the third people’s hospital of gansu province. All the specimens have been confirmed by pathological diagnosis. According to the international general diagnosis standards of cervical specimen, the specimen was devided into four groups:normal group (17 cases), CINI group (21 cases), CINⅡ-Ⅲ group (28 cases) and cervical squamous cell carcinoma group (30 cases). According to the standard of clinical stage of the international union of gynecology and obstetrics (FIGO) in 2009, the cervical squamous cell carcinoma group is divided into two groups:the Ⅰ-Ⅱ group(15 cases), Ⅲ-Ⅳ group(15 cases); According to the results of postoperative histopathologic examination and ultrasonic, CT, mri and other auxiliary examination to confirmed the existence of lymph node metastasis, the cervical squamous cell carcinoma group can be divided into two groups:lymph node metastasis group (12 cases), none lymph node metastasis group (18 cases); According to cellular histologic classification cervical squamous cell carcinoma group can be divided into three groups:high differentiated squamous cell carcinoma group (10 cases), medium differentiated squamous cell carcinoma group (9 cases), low differentiated squamous cell carcinoma group (11 cases); According to the average age (age 26~61, an average of about 40 years old) cervical squamous cell cancer group can be divided into two groups:younger than 40 years old group (13 cases), older than 40 years old group (17 cases). Experimental method:use immunohistochemical streptomycin avidin peroxidase-connecting method to test the positive expression rate of Bmi - 1 and PCNA of all the groups, and statistically analasis the clinical significance and relationship between cervical squamous cell carcinoma and Bmi-1 and PCNA.Results: the positive expression rate of Bmi-1 in the group of normal cervix, the cervix CINI group, CINⅡ-Ⅲ group and the cervical squamous cell carcinoma showing a increasing trend, respectively,11.8%,23.8%,50.0%,66.7%, the positive expression rate of cervical squamous cell cancer group was obviously higher than CIN group and cervical normal group, the positive expression rate of CIN group was obviously higher than that of normal group,the cervix CINⅡ-Ⅲ group obviously higher than that of CINI group and normal group, and comparative differences between groups was statistically significant (P< 0.05). The positive expression rate of PCNA in normal cervix, CINI group, CINⅡ-Ⅲ group and cervical squamous cell carcinoma group also showing a increasing trend respectively,17.6%,47.6%,82.1%,96.7%, the differences between groups was statistically significant (P< 0.05). The positive expression rate of Bmi - 1 in cervical squamous cell carcinoma group is associated with clinical stage (P< 0.05), and also associated with the presence of lymph node metastasis (P< 0.05), but has nothing to do with the pathologic stage and age (P> 0.05); the positive expression rate of PCNA in cervical squamous cell carcinoma group is associated with clinical stage, the presence of lymph node metastasis and pathologic stage (P<0.05),but has nothing to do with age(P>0.05) In cervical squamous cell carcinoma, the positive expression rate of Bmi-1 and PCNA has no correlation.Conclusion: in normal group, CINI group, CINⅡ-Ⅲ group and the cervical squamous cell carcinoma group, Bmi - 1 and PCNA expression showing a increasing trend, which prompts they are involved in the occurrence and development of the cervical squamous cell carcinoma, but the effect of Bmi - 1 and PCNA in the process of cervical squamous cell carcinoma seems independent. So the joint detection the expression of Bmi - 1 and PCNA in cervix can be an effective auxiliary means to diagnose and distinguish the severity and the prognosis about the cervical lesions.
