BackgroundTo compare the laparotomy,laparoscopic operation has less pain,length of stay and become important way to treat innocent gynecological diseases.With the development of laparoscopic operation,it's application in women's malignant tumour ,for example, early stage of ovarian cancer, endometrial cancer and endometrial cancer develope too.Recently, more and more people pay attention to the application of laparoscope in women's malignant tumor .But the arguments about the safety of the operation is still very intense,especially whether carbon dioxide can promote the development and metastasis of tumor cells.It's reported that carbon dioxide can alter the biologic activity of the tumor cells , prompt the tumor growth and cause metastasis on the puncture and dissemination through abdominal cavity.There's also adverse report , Canis made use of rat tumor model to explore the impacts of carbon dioxide pneumoperitoneum to tumor ,the results suggestted bigger tumor volume in laparotomy and more tumor spread in laparoscopic operation.But the mechanisms are still uncertain .There were various theories in nation and abroad.For example, acidosis , immunologic injury, physical damage et al.At present,reports about transfer agent and adhesion molecule are still rare. Section I Effects of carbon dioxide on the expressions of XIAP and PED/PEA-15 of human ovarian cancer cell line SKOV3Objective: To explore the effects of carbon dioxide on the expressions of antiapoptosis factors XIAP and PED/PEA-15 of human ovarian cancer cell line SKOV3.Methods: Put human ovarian cancer cell line SKOV3 in analogous CO2 pneumoperitoneum environment.Realtime FQ-PCR and Immunocytochemical staining were used to examine the expressions of XIAP and PED/PEA-15 of these cells after exposure to different pressure and persist time of CO2.Results: CO2 could significantly increase the expressions of XIAP and PED/PEA-15(P<0.05).The extent was related with the pressure and persistence time of CO2.Conclusion: CO2 can suppress apoptosis capacity of SKOV3 cell by upregulating the expressions of antiapoptosis factors XIAP and PED/PEA-15.Section II Effects of CO2 on the adhesion and metastasis of human ovariancarcinoma cell line SKOV3Objective: To explore the effects and the related mechanism of CO2 on the adhesion and metastasis of human ovarian carcinoma cell line SKOV3.Methods: SKOV3 was cultured in analogous CO2 pneumoperitoneum environment. Cell adhesion capacity was determined after exposure to different pressure and time of CO2. Realtime FQ-PCR and Immunocytochemical staining was used to examine the expression of CD44v6, MMP-9 and E-cad of these cells.Results: CO2 could enhance SKOV3 cell's adhesion in a pressure and time dependent fashion, and the peak amplitude was in the combined effect of 10mmHg,2h and 48h. CO2 could slightly downregulate the expression of E-cad , significantly upgrade the expression of CD44v6 and MMP-9.The combined effects of pressure and time were more striking in changing the expression of CD44v6 and MMP-9.Conclusion: CO2 could increase the cell adhesion capacity and promote tumor metastasis through upregulating expressions of CD44v6 and MMP-9, even downregulating E-cad expression. The combination of pressure and time could manifest a stronger effect on the adhesion and metastasis of SKOV3.
