Background and Objective: Since Starzal initiating the first case of authotopic liver transplantation in 1963, as the improvement of liver transplantation surgical technic, and the widely clinical application of cyclosporim, FK-506, UW solution, the long-term survival rate after liver transplantation increased tremendously. Liver transplantation has become an operative therapeutic approach to cure acute and chronic liver function failure caused by final-stage cirrhosis, liver malignant tumor, inborn and metabolic diseases, etc. At present, many factors restricted the clinic results of liver transplantation. The quality of donor, as the key element in liver transplantation, has aroused general concern. The most objective index to test the quality of donor is "zero point liver biopsy". The aim of this experiment is to observe cell apoptosis after donor hypothermal conservation by applying "zero point liver biopsy" method.Methods: Procurement 30 donors by using normal position by-pass perfusion procurement method, keep them in UW solution at 0-4°C, divided them into 3 groups according to their preserved time. At the end of their preserved term, take 0.5~*0.5~*0.5cm liver tissue at the margin of donor's left lobe, fix the tissue by 4% formalin, paraffin imbedding, make serial sections, and at last, observe serial section's apoptosis phenomenon by TUNEL (terminal deoxynucleotidyl transferase mediated UTP-FITC nick end labeling) means, count the apoptosis cell, and calculate the apoptosis index.Resule one: The 30 liver donor was procured integrated. There was no damnificationon the vein. The liver donor was perfused well and the liver function resumed well.Resule two: In the three groups the positive cell is: 1.20±0.82,4.06±1.09,6.98±2.25.The apoptosis index is: 0.86±0.77, 3.26±1.10, 5.86±1.97.Conclusion: Cell apoptosis exists at the end of donor liver hypothermal preservedterm, and it is a direct ratio between the apoptosis degree and lasting time ofhypothermal preserved term.
