Abstract/Summary:
Objectiv:To investigate the effect and mechanism of low density lipoprotein on multiple organ dysfunction in endotoxemia rats. Methods:Endotoxemia was induced by a slow intravenous injection of 5mg/kg lippolysaccharide (Escherichia coli 055 ? B5) via tail vein of rats for 20min. 60 male Wister rats were randomly divided into three groups as follows:normal control group (n=18), endotoxemia group(n=18) and low density lipoprotein pretreatment group (n=18) , every group further divided into 1.5hours, 6hours and 12hours subgroup, 6 rats for each subgroup. At the three time points, rats in all groups were sacrificed meanwhile blood and tissue sample from lungs and small investine were harvested to determine serum aspatate transferase (AST), alanine transferase (ALT), blood urea nitrogen (BUN) , creatinine(Cr), tumor necrosis factor-α (TNF-α)and interleukin-6(IL-6), pulmonary myeloperoxidase activities (MPO) and small intestinal diamine oxidase(DAO) activities. Results:In endotoxemia rats, serum TNF-α concentration significantly increased at 1.5hours (P<0.01) and was still higher than normal control at 6 hours (P<0.05), then restored to the control at 12hours. Serum IL-6 concentration reached the peak at 6hours (P<0.01) and remained higher than normal control at 12hours(P<0.01). Serum AST (P<0.01) as well as ALT(P<0.05) levels and Cr(P<0.05) levels at 12 hours in the endotoxemia group were much higher than those in normal control.
