Objective:Sialidase is a kind of glycosidase, which can hydrolyze sialic acid from non-deoxidized endglycogen and glycolipid. Human sialidases are classified into four types, among which human membrane-associated sialidase (hmSD) is more important. HmSD is unique in specifically hydrolyzing gangliosides, participates in cell differentiation and transmembrane signaling, and plays crucial roles in the regulation of cell functions. Many recent studies indicate that high expression of hmSD in cancer cells leads to protection from programmed cell death, probably due to the capability of regulating gangliosides. Therefore further investigation may provide a possible sialidase target for diagnosis and therapy of prostatic cancer. In this study, PC3-hmSD cell line established with stablely transfected hmSD and PC3 exposes to VK3 or NaBT,then analyzes the relationships between hmSD activity and apoptotic rate, and apoptosis related pathway (oxidative stress, NF-κB, caspase9, bcl-xl). The aim of this study is to discuss the mechanisms of hmSD against apoptosis, and this is helpful to the clinical diagnosis and therapy of prostatic cancer.
